I was a strong mission runner able to grind thru lvl4s in rapid succession. I went tropo, moved into a C2 w-system, and haven't looked back. I upgraded to a C3 once the C2 sites became boringly easy, and have recently moved on to a C4 w-system for similar reasons. My C4 income atm is less than when I was in the C3 but is still way way more than I was ever able to make running missions.
A previous responder, however, summed it up nicely with an ever applicable "it depends".
Living safely and profitably in w-space requires quite a bit of effort, and you need a lot of skills (both skillbook and gameplay) that you do not need to run missions. A mission-runner can ignore their play for a week or two, and safely resume where they left off. A w-space resident cannot really afford to do this ... as a number of exam-time POS owners have discovered over recent weeks. So if you're a sporadic player then w-space residence may not be best for you. However, if you're the member of a large and active w-space corp then you may be able to get by despite being a sporadic player.
When working in my C3 several hundred million isk per hour of sleeper killing was the norm, and a billion or two for a busy session was not unusual. The catch was that, unlike missions, there is no ready and infinite supply of w-space sigs and anomalies. If you clear what you have in your system then you must wait, sometimes for a long period, for new sites to spawn. If you're raiding neighbouring systems, and have cleared the system, then you need to collapse the adjoining WH and checkout the new WH and system.
Back in the 'it depends' basket is the number of people you're working with. If you're a regular player then solo-running a C2, C3, or C4 will make you heaps more isk (skill for skill, hour for hour) than mission running ... more so if you're harvesting all the resources the system offers you. I have no experience with C1s but suspect they would also offer more than mission-running. If you're part of a large group in a w-system then you, individually, may not be able to make as much as you would running missions ... it just depends on how many people each get a share of the w-system takings.
Best w-space income? Probably either solo (or very small group) living-in and running a C2 or C3 system (possibly C4 but I'm yet to experience that) OR as part of a larger corp that raids through its static WHs for most of its income. Of course your income is better if it's not entirely dependent on combat sites, but also supplemented by ladar / grav sigs and PI.
I operate my w-space efforts very differently to Mr Kidd but, just like him, I am making heaps more isk than I ever did as a skilled hard-working mission runner. Profit!
